World’s best C&G Stain

The detection of Cryptosporidium and Giardia in water is extremely difficult. There are many particles present in water samples that cross-react with Cryptosporidium-specific and Giardia-specific antibodies. These particles can easily be mistaken for Cryptosporidium oocysts or Giardia cysts.

EasyStain™ is an immunofluorescence reagent designed for use in testing water samples for the presence of Cryptosporidium oocysts and Giardia cysts. When a water sample is stained with the Cryptosporidium and Giardia  reagent, all Cryptosporidium oocysts or Giardia cysts present in the sample will fluoresce bright green. Stained samples can then be examined using fluorescence microscopy.

Unlike other Cryptosporidium and Giardia antibodies, EasyStain™ is revolutionary in that it produces minimal background fluorescence. It is sold as a kit containing sufficient reagents to perform 80 tests. The kit shelf life is 12 months from date of manufacture.

Tips for US EPA Method 1623 Users

Here are some tips for using EasyStain™ in US EPA Method 1623 that we’d like to share with EasyStain™ users.

Slide Drying

Method 1623 does not state the time that labs can take to dry slides after preparation. To obtain consistent staining with EasyStain™ in Method 1623, slides must be dried rapidly at 37° C and stained immediately. Alternatively slides can be dried overnight in a refrigerator.

If slides are not dried quickly, inconsistent staining will result despite how the sample is stained afterwards.

Slides may not dry in cold rooms since some cold rooms maintain a humid environment. Slides usually dry within 4 hours in a standard refrigerator.

DAPI Staining

A methanol fixation step is included in the EasyStain™ instructions to assist permeabilization of oocysts and cysts prior to DAPI staining. Some people have reported that the methanol step improves DAPI staining, whilst others have reported that it has not. 

The methanol fixation step of the EasyStain™ method can be left out at the discretion of the laboratory.
